dc.description.abstract | [ES] El Programmed Death-1 Homolog (PD-1H) es una molécula perteneciente a la familia de CD28/B7 de receptores de membrana. Su función es la de inhibir la activación linfocitaria tras la interacción con su ligando y debido a esto, está considerado como una diana terapéutica muy prometedora en inmunoterapia del cáncer. Sin embargo, en la actualidad no se conoce con seguridad cual es el ligando de este receptor a pesar de varios estudios que han logrado identificar interacciones de PD-1H con otras moléculas. En este trabajo se describe la puesta a punto de un método experimental para la expresión de la proteína recombinante de fusión PD1H/BioID2. Concretamente, se ha generado una construcción genética recombinante de una fusión entre el receptor celular PD-1H y la biotin ligasa BioID2, capaz de marcar proteínas con biotina por proximidad. Utilizando esta construcción, se modificaron genéticamente células con el objetivo de aislar posteriormente sus proteínas y purificar aquellas marcadas con biotina. Asimismo, se han preparado extractos de lisados celulares a partir de células transducidas que expresan dicha proteína recombinante de fusión para identificar los posibles ligandos que se hayan biotinilado de PD-1H | es_ES |
dc.description.abstract | [EN] Programmed Death-1 Homolog (PD-1H) is a molecule belonging to the CD28/B7 family of membrane receptors. Its function is to inhibit lymphocyte activation upon interaction with its ligand and because of that, it is considered a very promising therapeutic target in cancer immunotherapy. However, nowadays, the ligand of this receptor is not known with certainty despite the fact that several studies have identified interactions of PD-1H with other molecules. This work describes the development of an experimental method for the expression of the recombinant PD-1H/BioID2 fusion protein. Specifically, a recombinant genetic construct of a fusion between the cellular PD-1H receptor and the biotin ligase BioID2, capable of tagging biotin-binding proteins by proximity, has been generated. Using this genetic construct, cells were genetically modified in order to subsequently isolate biotin-tagged proteins.
